Which extension ?
for which type of website ?
Under any Windows OS, the extension can be choosen in the "Save as" window, in the bottom dialog box.

1 ) First and important
If you want a chance to edit your image later save always your work as .pfi, so you will save layers and alike

2) To upload on the web use jpg...try to use GIF only for animation because gif quality is very low...(as example support only 255 colours instead of the millions of other format...and that is only 1 of its disadvantage)

If your image contain transparency is better use PNG that GIF, Quality of png is much higher and what is more important support PARTIAL transparency

Only issue of Png on the web was with old version of INTERNET EXPLORER that didn't display correctly its transparence, BUT this was fixed with last version

So basically

1 normal image jpg
2 image with transparent areas png
3 ANimation gif

ORIGINAL (if has more then 1 layer)ALWAYS MUST SAVED AS PFI, and then save a copy for the web
---------------------------------------------------------------------------

Mind that jpg is a lossy format,...that means that even saving with no compression, ANYTIME you open and resave a jpg you loose a bit of quality
