no, middle click not supported, only middle mouse wheel for scrolling

Learn to use the keyboard shortcuts instead.
The shortcut for Hand tool is U, so simply typing U on the keyboard will get the tool automatically. The if for example you then need the brush tool simply type P. This is a simpler way and its how the pro's do it.

Can you change the keyboard shortcuts to anything you like or are they locked?

In Photofiltre 7 they are pretty much locked.
